Differences in trait impulsivity do not bias the response to pharmacological drug challenge in the rat five-choice serial reaction time task.
RATIONALE: Maladaptive impulsivity is symptomatic of several neuropsychiatric disorders including schizophrenia, attention-deficit hyperactivity disorder (ADHD), and substance abuse disorders; paradigms designed to assess the underlying neurobiology of this behavior are essential for the discovery of novel therapeutic agents. Various models may be used to assess impulsivity as measured by the five-choice serial reaction time task (5-CSRTT), including variable inter-trial interval (ITI) sessions, the selection of extreme high and low impulsivity phenotypes from a large outbred population of rats, as well as pharmacological challenges. OBJECTIVES: The aim of this study is to evaluate if pharmacological challenge models for impulsivity are biased by underlying differences in impulsivity phenotype. METHODS: Extreme high and low impulsivity phenotypes were selected in the 5-CSRTT, and dose-dependent effects of various pharmacological challenges, namely MK-801, yohimbine, and cocaine, were evaluated on task performance, specifically accuracy and premature responses. RESULTS: All three compounds increased premature responding, while a decrease in attentional performance occurred following MK-801 and yohimbine administration. No differences in drug-induced impulsivity between rats selected for high or low impulsivity or in parameters indicative of attentional performance could be determined. CONCLUSIONS: Our findings indicate that different pharmacological challenges increase impulsivity on the 5-CSRTT, with modest effects on attention. These effects were not influenced by underlying differences in impulsivity phenotype, which is an important prerequisite to reliably use these challenge models to screen and profile compounds with putative anti-impulsive characteristics
A Gene Expression Fingerprint of C. Elegans Embryonic Motor Neurons
Differential gene expression specifies the highly diverse cell types that constitute the nervous system. With its sequenced genome and simple, well-defined neuroanatomy, the nematode C. elegans is a useful model system in which to correlate gene expression with neuron identity. The UNC-4 transcription factor is expressed in thirteen embryonic motor neurons where it specifies axonal morphology and synaptic function. These cells can be marked with an unc-4::GFP reporter transgene. Here we describe a powerful strategy, Micro-Array Profiling of C. elegans cells (MAPCeL), and confirm that this approach provides a comprehensive gene expression profile of unc-4::GFP motor neurons in vivo.
The Wyoming Survey for H-alpha. I. Initial Results at z ~ 0.16 and 0.24
The Wyoming Survey for H-alpha, or WySH, is a large-area, ground-based,
narrowband imaging survey for H-alpha-emitting galaxies over the latter half of
the age of the Universe. The survey spans several square degrees in a set of
fields of low Galactic cirrus emission. The observing program focuses on
multiple dz~0.02 epochs from z~0.16 to z~0.81 down to a uniform
(continuum+line) luminosity at each epoch of ~10^33 W uncorrected for
extinction (3sigma for a 3" diameter aperture). First results are presented
here for 98+208 galaxies observed over approximately 2 square degrees at
redshifts z~0.16 and 0.24, including preliminary luminosity functions at these
two epochs. These data clearly show an evolution with lookback time in the
volume-averaged cosmic star formation rate. Integrals of Schechter fits to the
extinction-corrected H-alpha luminosity functions indicate star formation rates
per co-moving volume of 0.009 and 0.014 h_70 M_sun/yr/Mpc^3 at z~0.16 and 0.24,
respectively. The formal uncertainties in the Schechter fits, based on this
initial subset of the survey, correspond to uncertainties in the cosmic star
formation rate density at the >~40% level; the tentative uncertainty due to
cosmic variance is 25%, estimated from separately carrying out the analysis on
data from the first two fields with substantial datasets.Comment: To appear in the Astronomical Journa
The Lilead Survey: A National Study of District-Level Library Supervisors: Roles, Responsibilities, Challenges, and Professional Development Needs
The school district library supervisor plays a pivotal role in supporting, advising, and providing professional development to building-level librarians; advocating for the program; providing leadership; and representing school library programs to stakeholders in the school system and the larger community. To gain a better understanding of supervisors\u27 roles, responsibilities, demographics, and challenges, and to establish baseline data upon which further research can be built, the Lilead Project was initiated in 2011 at the University of Maryland with funding from the Institute of Museum and Library Services. In 2012 the project team conducted the Lilead Survey, a survey of supervisors nationwide. In this paper, the second of two reports on the results of the survey, we present findings related to the responsibilities and tasks assigned to the position, professional development needs of supervisors and staff, and the range of stakeholder groups with which supervisors work
The Lilead Survey: A National Study of District-Level Library Supervisors: The Position, Office, and Characteristics of the Supervisor
The school district library supervisor occupies a pivotal position in library and information services programs that support and enhance the instructional efforts of a school district: providing leadership; advocating for the programs; supporting, advising, and providing professional development to building-level librarians; and representing school library programs to stakeholders in the school system and the community at large. With funding from the Institute of Museum and Library Services, the Lilead Project was founded at the University of Maryland in 2011 to study, support, and build community among school district library supervisors (Lilead Project n.d.). To gain a better understanding of supervisors--who they are, the duties they perform, and the challenges they face--and to establish baseline data upon which further research can be built, in 2012 the project team conducted a survey of supervisors nationwide: the Lilead Survey. In this paper, which is the first of two planned reports on the results of the survey, we present findings related to the position and office of the supervisor; demographic information, qualifications, and career paths of the incumbents of the position; and changes in policies, curriculum, and resources that impact the supervisor\u27s responsibilities for library services. Survey findings related to responsibilities and tasks assigned to the position, professional development needs of supervisors and staff, and challenges and needs that supervisors face will be presented in the second report

Ventral Striatal D2/3 Receptor Availability Is Associated with Impulsive Choice Behavior As Well As Limbic Corticostriatal Connectivity.
BACKGROUND: Low dopamine D2/3 receptor availability in the nucleus accumbens shell is associated with highly impulsive behavior in rats as measured by premature responses in a cued attentional task. However, it is unclear whether dopamine D2/3 receptor availability in the nucleus accumbens is equally linked to intolerance for delayed rewards, a related form of impulsivity. METHODS: We investigated the relationship between D2/3 receptor availability in the nucleus accumbens and impulsivity in a delay-discounting task where animals must choose between immediate, small-magnitude rewards and delayed, larger-magnitude rewards. Corticostriatal D2/3 receptor availability was measured in rats stratified for high and low impulsivity using in vivo [18F]fallypride positron emission tomography and ex vivo [3H]raclopride autoradiography. Resting-state functional connectivity in limbic corticostriatal networks was also assessed using fMRI. RESULTS: Delay-discounting task impulsivity was inversely related to D2/3 receptor availability in the nucleus accumbens core but not the dorsal striatum, with higher D2/3 binding in the nucleus accumbens shell of high-impulsive rats compared with low-impulsive rats. D2/3 receptor availability was associated with stronger connectivity between the cingulate cortex and hippocampus of high- vs low-impulsive rats. CONCLUSIONS: We conclude that delay-discounting task impulsivity is associated with low D2/3 receptor binding in the nucleus accumbens core. Thus, two related forms of waiting impulsivity-premature responding and delay intolerance in a delay-of-reward task-implicate an involvement of D2/3 receptor availability in the nucleus accumbens shell and core, respectively. This dissociation may be causal or consequential to enhanced functional connectivity of limbic brain circuitry and hold relevance for attention-deficit/hyperactivity disorder, drug addiction, and other psychiatric disorders
Prolyl-4-hydroxylase 3 maintains β-cell glucose metabolism during fatty acid excess in mice
The α-ketoglutarate–dependent dioxygenase, prolyl-4-hydroxylase 3 (PHD3), is an HIF target that uses molecular oxygen to hydroxylate peptidyl prolyl residues. Although PHD3 has been reported to influence cancer cell metabolism and liver insulin sensitivity, relatively little is known about the effects of this highly conserved enzyme in insulin-secreting β cells in vivo. Here, we show that the deletion of PHD3 specifically in β cells (βPHD3KO) was associated with impaired glucose homeostasis in mice fed a high-fat diet. In the early stages of dietary fat excess, βPHD3KO islets energetically rewired, leading to defects in the management of pyruvate fate and a shift from glycolysis to increased fatty acid oxidation (FAO). However, under more prolonged metabolic stress, this switch to preferential FAO in βPHD3KO islets was associated with impaired glucose-stimulated ATP/ADP rises, Ca(2+) fluxes, and insulin secretion. Thus, PHD3 might be a pivotal component of the β cell glucose metabolism machinery in mice by suppressing the use of fatty acids as a primary fuel source during the early phases of metabolic stress
Investigating hyper-vigilance for social threat of lonely children
The hypothesis that lonely children show hypervigilance for social threat was examined in a series of three studies that employed different methods including advanced eye-tracking technology. Hypervigilance for social threat was operationalized as hostility to ambiguously motivated social exclusion in a variation of the hostile attribution paradigm (Study 1), scores on the Children’s Rejection-Sensitivity Questionnaire (Study 2), and visual attention to socially rejecting stimuli (Study 3). The participants were 185 children (11 years-7 months to 12 years-6 months), 248 children (9 years-4 months to 11 years-8 months) and 140 children (8 years-10 months to 12 years-10 months) in the three studies, respectively. Regression analyses showed that, with depressive symptoms covaried, there were quadratic relations between loneliness and these different measures of hypervigilance to social threat. As hypothesized, only children in the upper range of loneliness demonstrated elevated hostility to ambiguously motivated social exclusion, higher scores on the rejection sensitivity questionnaire, and disengagement difficulties when viewing socially rejecting stimuli. The Wyoming Survey for H-alpha. III. A Multi-wavelength Look at Attenuation by Dust in Galaxies out to z~0.4
We report results from the Wyoming Survey for H-alpha (WySH), a comprehensive
four-square degree survey to probe the evolution of star-forming galaxies over
the latter half of the age of the Universe. We have supplemented the H-alpha
data from WySH with infrared data from the Spitzer Wide-area Infrared
Extragalactic (SWIRE) Survey and ultraviolet data from the Galaxy Evolution
Explorer (GALEX) Deep Imaging Survey. This dataset provides a multi-wavelength
look at the evolution of the attenuation by dust, and here we compare a
traditional measure of dust attenuation (L(TIR)/L(FUV)) to a diagnostic based
on a recently-developed robust star formation rate (SFR) indicator,
[H-alpha_obs+24-micron]/H-alpha_obs. With such data over multiple epochs, the
evolution in the attenuation by dust with redshift can be assessed. We present
results from the ELAIS-N1 and Lockman Hole regions at z~0.16, 0.24, 0.32 and
0.40. While the ensemble averages of both diagnostics are relatively constant
from epoch to epoch, each epoch individually exhibits a larger attenuation by
dust for higher star formation rates. Hence, an epoch to epoch comparison at a
fixed star formation rate suggests a mild decrease in dust attenuation with
